Overview

In der vorliegenden Studie wurde Sporotrichum thermophile ATCC 42464 für die Produktion von Serinprotease mittels Feststoffsubstratfermentation (SSF) verwendet. Weizenkleie (12,5 g) wurde als Kohlenstoffquelle eingesetzt. Die Kulturbedingungen wie Feuchtigkeitsgehalt (MSS-12,5 ml), pH-Wert (5) und Inkubationszeit (48 Stunden) wurden optimiert. Die Enzymaktivität nahm in Gegenwart von Inhibitoren (Harnstoff, SDS) und Chelatoren (EDTA) ab, während die Anwesenheit von Metallionen, insbesondere Mg2+, eine Zunahme der Aktivität der Serinprotease verursachte. Die maximale Enzymaktivität von 18,84 U/g wurde unter den optimierten Bedingungen beobachtet.
