OverviewOver the last 10 years, it has become apparent that poly-ADP ribosylation reactions govern some of the fundamental processes of life. PARP 1 is a highly conserved enzyme and a constitutive factor of the DNA damage surveillance network and plays an essential role in the protection and processing of DNA ends as they arise in DNA damage, repair, and recombination. The recent discovery of multiple members of the PARP family suggests an expanded role for poly-ADP ribosylation reactions. Poly ADP-ribosylation Reactions contains basic knowledge of poly-ADP ribosylation reactions and reviews the most recent molecular (including the crystal structure) and genetic (including knock out mice) approaches that have contributed to our understanding of the physiological role PARP 1 and its homologues. A discussion of the novel pharmacological role of inhibition of PARP is also included. The final chapter outlines future directions in the field of cellular responses to DNA damage. Its comprehensive coverage of the multiple aspects of this type of post-translational modification induced by DNA damage mean this book will serve as reference for both new and experienced investigators. Trends in Genetics From DNA Damage and Stress Signalling to Cell Death: Poly(ADP-ribosyl)ation Reactions is the first monograph fully dedicated to the discussion of the biochemistry, molecular biology and molecular genetics of protein-poly(ADP-ribosyl)ation reactions. The literature cited in this monograph encompasses work that began in the 60s with the discovery of the natural occurrence of ADP-ribose polymers, continuing until 1999. ... The book is well written and organized, and is equally useful for the experts and for scientists just entering this exciting and rapidly moving field. The monograph starts with a discussion of ADP-ribose polymer metabolism and continues with the structural characterization of PARP-1 at the molecular level. It also elegantly describes the application of modern molecular and genetic approaches to the elucidation of the biological significance of protein-poly(ADP-ribosyl)ation pathways. --Trends in Genetics<br> Author InformationTab Content 6Author Website:Countries AvailableAll regions |
