Abstract: Abstract of thesis entitled Investigation on the Differential Expression and Hormonal Regulation of Olfactomedin in Uterus Submitted by Pak-Yiu NG for the Degree of Master of Philosophy at The University of Hong Kong in August 2007 Introduction: Recent microarray studies in humans showed that endometrial Olfactomedin (Olfm) is down-regulated during the receptive period when compared with the non-receptive periods. Yet, the underlying molecular mechanisms that control Olfm-1 expression and its role on the implantation process are largely unknown. The objectives of this study were to investigate the differential expression of Olfm in human and mouse uterus and to study the effect of ovarian hormones on Olfm expression in vitro and in vitro models. Materials and Methods: Endometrial Olfm expression and localization were studied by immunohistochemistry and quantitative PCR. The effect of estrogen (E2) and progesterone (P4) on Olfm regulation was studied using in-vitro human primary endometrial, ovariectomized and delayed implantation models. Results: Human endometrial Olfm-1 and Olfm-2 transcripts decreased significantly from the proliferative to the secretory phases of the menstrual cycle and in stimulated IVF patients with high serum E2 levels (>=20,000 pmol/L) than the natural cycle controls. Olfm-1 transcript and protein are highly expressed in human endometrial epithelium. However, Olfm-1 transcript was stimulated by E2 and P4 treatment in human primary endometrial cells. In mouse, Olfm-1 transcript is transiently suppressed at 3 hours post-estrogen injection in delayed implantation and E2-treated ovariectomized models. Conclusions: Olfm is differentially regulated by steroid hormones during the implantation window and down-regulation of Olfm may play an important role on the implantation process in humans and mice.
