OverviewPoor water solubility of active pharmaceutical ingredients (APIs) presents a significant challenge in oral drug administration, leading to reduced bioavailability and compromised therapeutic effectiveness. This study aimed to address this issue by developing an optimized itraconazole nanosuspension for intranasal delivery, potentially enhancing drug availability across the blood-brain barrier. Then a nanosuspension was prepared and characterized using various analytical techniques. FTIR and DSC analyses verified the drug's chemical structure and crystalline state .A Box-Behnken Design was employed to optimize the formulation, resulting in nanoparticles with favorable size, polydispersity index, zeta potential, and entrapment efficiency. TEM imaging revealed a core-shell structure of the nanoparticles. The optimized nanosuspension demonstrated improved drug release compared to a conventional suspension. These findings suggest that the developed itraconazole nanosuspension has potential to enhance the drug's solubility and bioavailability.
