Pedro Ponce estudio en el Colegio de Santa Cruz en Mexico. Se graduo como licenciado en teologia y, entre 1571 y 1626, tuvo a su cargo la diocesis de Zumpahuacan. Una cronica de la epoca relata que: En 1565 fue construido el templo parroquial para la congregacion de los Agustinos ... por orden de Pedro Ponce de Leon, probablemente hijo de Lucas Ponce de Leon, hijo del rey Cuatlatlapatzin de Tlaxcala.
